I ran an ad in the newspaper announcing 'Second Printing Ready.'

Literal

Newspaper [in-に] 'reprint-available' [quotative-と] advertised.

Publishing industry jargon. 重版出来 (じゅうはん しゅったい) is a set phrase specifically used in publishing to announce that a reprint of a book is ready to ship — i.e. the book sold out the first run and is getting a second printing. It's traditionally used as an announcement in newspaper ads or on obi. 出来 here is read 'しゅったい' ('completion, ready to come out'), not 'でき.'
